本研究采用形态学观察结合核糖体ITS、叶绿体rbcL基因和5S rDNA间隔序列3个分子标记系统进化分析的研究方法,对来自宁波梅山港的进境船舶压舱水中的一株石莼属绿藻进行了种类鉴定。形态学观察结果表明其具有藻体中空管状、具分枝、中部细胞呈典型纵向排列细胞、无明显的圆角和帽状叶绿体、每个细胞中有蛋白核1个到2个,少数细胞具有3个蛋白核的形态特征。ITS和rbcL序列分析结果均将其划分在LPP分枝,5S rDNA间隔序列分析表明该绿藻属于U.prolifera分枝。综合以上结果,将其鉴定为浒苔U.prolifera,这是国内外首次报道浒苔可通过压舱水进行传播的报道,也是我国首次从进境压舱水中截获浒苔这一绿潮藻。
In this study, morphological observation combined with ribosomal ITS, chloroplast rbcL gene and 5S rDNA spacer sequence analysis of three molecular marker system research methods, from Ningbo Meishan Port entry ship ballast water of a genus of Lithocarpus The type of identification. ITS and rbcL sequence analysis results are divided into LPP branches, 5S rDNA interval sequence analysis showed that the green algae belong to U. prolifera branch. Based on the above results, it was identified as Enteromorpha U. prolifera, which is the first report that Enteromorpha can be transmitted by ballast water at home and abroad. It is also the first time that China captured Enterolomium, a green tide algae, from the entry ballast water.
