目的观察重楼皂苷Ⅰ对人肝癌细胞MHCC97-H增殖、周期及凋亡的影响。方法 1.将对数生长期的人肝癌细胞MHCC97-H,加入不同浓度的重楼皂苷Ⅰ和索拉菲尼,24h后CCK8法检测药物对细胞增殖的影响。2.根据CCK8实验结果,选取相应浓度的重楼皂苷Ⅰ和索拉菲尼,作用于细胞24h,观察细胞周期及凋亡情况。结果重楼皂苷Ⅰ及索拉非尼作用于人肝癌细胞MHCC97-H的IC50的浓度分别是5.7μg/m L、10μmol/m L。重楼皂苷Ⅰ及索拉菲尼均能抑制S期,将细胞阻滞于G0/G1期,能明显促进细胞凋亡,与空白对照组比较具有显著差异(P<0.01)。结论重楼皂苷Ⅰ对人肝癌细胞MHCC97-H具有明显的抑制增殖及促进凋亡的作用。
Objective To observe the effect of polysaccharide Saponins Ⅰ on the proliferation, cell cycle and apoptosis of human hepatoma cell line MHCC97-H. Methods 1. The logarithmic growth phase of human hepatocellular carcinoma cells MHCC97-H, adding different concentrations of polysaccharide saponin Ⅰ and sorafenib, CCK8 24h detection of drug effects on cell proliferation. According to CCK8 experimental results, select the corresponding concentrations of polysaccharide saponin Ⅰ and sorafenib, the role of cells 24h, observe the cell cycle and apoptosis. Results The concentrations of IC50 of resplenverting Ⅰ and sorafenib in MHCC97-H cells were 5.7 μg / ml and 10 μmol / ml, respectively. Saponin Ⅰ and sorafenib can inhibit S phase, the cells arrested in the G0 / G1 phase, can significantly promote apoptosis, compared with the control group was significantly different (P <0.01). Conclusions Saponin Ⅰ has obvious inhibitory effect on proliferation and apoptosis of human hepatoma cell line MHCC97-H.
