目的探讨儿茶酚胺氧位甲基转移酶(catechol O-methyltransferase,COMT)m RNA及其蛋白在结直肠腺瘤及其瘤旁组织、结直肠癌(CRC)及其癌旁组织中的表达情况。方法采用实时定量PCR和免疫组化方法检测结直肠腺瘤及其瘤旁组织、CRC及其癌旁组织中COMT m RNA及其蛋白的表达情况,并探索COMT表达与结直肠腺瘤和CRC患者临床病理特征的关系。结果 1实时定量PCR结果显示:结直肠腺瘤组织中COMT m RNA表达水平的中位数为0.109 0,高于瘤旁组织的0.000 5(t=3.02,P=0.01);CRC组织中COMT m RNA表达水平的中位数为0.041 8,高于其癌旁组织的0.013 5(t=2.71,P=0.02)。2免疫组化染色结果显示:结直肠腺瘤组织中,COMT的高表达率为72.34%(34/47),高于瘤旁组织的25.53%(12/47),χ2=28.72,P<0.01;CRC组织中COMT的高表达率为66.67%(28/42),高于癌旁组织的28.57%(12/42),χ2=4.97,P<0.05。3在结直肠腺瘤组织中,COMT高表达与患者的性别、年龄、肿瘤部位及病理学类型均无关(P>0.05);在CRC组织中,COMT高表达与患者的性别、年龄、肿瘤部位及分化程度均无关(P>0.05),但COMT高表达与TNM分期、T分期及N分期均相关(P<0.05),TNMⅠ+Ⅱ期、T1+T2期及N0期患者的COMT高表达率较高。结论 COMT在结直肠腺瘤及CRC组织中的表达较相应的瘤(癌)旁组织均升高,其可能在CRC的发生和发展中具有一定作用。
Objective To investigate the expression of catechol O-methyltransferase (mtna) m RNA and its protein in colorectal adenoma and its adjacent tissues, colorectal cancer (CRC) and its adjacent tissues. The results of immunohistochemical staining showed that the high expression rate of COMT was 72.34% (34/47) in colorectal adenoma tissues, which was higher than that in adjacent tissues (25.53%, 12/47), χ2 = 28.72, P <0.01 ; The high expression rate of COMT in CRC tissues was 66.67% (28/42), which was higher than that in paracancerous tissues (28.57%, 12/42), χ2 = 4.97, P <0.05.3 In colorectal adenoma tissues, COMT There was no correlation between the high expression of COMT and the gender, age, tumor location and pathological type (P> 0.05). The high expression of COMT had no correlation with gender, age, tumor location and differentiation (P> 0.05) , But the high expression of COMT was correlated with TNM stage, T stage and N stage (P <0.05). The high expression rate of COMT in TNMⅠ + Ⅱ stage, T1 + T2 stage and N0 stage patients was higher. Conclusion The expression of COMT in colorectal adenoma and CRC tissues is higher than that in the corresponding tumor tissues, which may play a role in the occurrence and development of CRC.
