目的探讨基质金属蛋白酶(MMP)-2、MMP-13在风湿性心脏病(RHD)二尖瓣组织中的表达和作用。方法实验组为成人RHD患者16例,对照组为无心脏病变的成人意外死亡者7例。经HE染色及电镜观察两组组织学及超微结构变化,采用免疫组织化学染色(SABC法)测定两组二尖瓣组织中MMP-2、MMP-13的表达情况。结果实验组为风湿性病理改变,对照组结构基本正常。两组比较MMP-2、MMP-13的积分光密度(IOD)Z值分别为-2.633,-2.936,差异均有统计学意义(P<0.01)。结论MMP-2、MMP-13的表达上调可能参与二尖瓣细胞外基质的重构,是其风湿性改变的重要分子机制之一。
Objective To investigate the expression and role of matrix metalloproteinase (MMP) -2 and MMP-13 in mitral valve tissue of rheumatic heart disease (RHD). Methods The experimental group was 16 adults with adult RHD and the control group was 7 adults without accidental heart disease. The histological and ultrastructural changes of the two groups were observed by HE staining and electron microscopy. The expression of MMP-2 and MMP-13 in mitral valve tissue was detected by immunohistochemical staining (SABC method). Results The experimental group was rheumatic pathological changes, the structure of the control group was normal. The integral optical density (IOD) Z values of MMP-2 and MMP-13 in the two groups were -2.633 and -2.936, respectively, with statistical significance (P <0.01). Conclusion The up-regulation of MMP-2 and MMP-13 may be involved in the remodeling of mitochondrial extracellular matrix, which is one of the important molecular mechanisms of rheumatic changes.
