用硫氰酸钾盐析法二次纯化人脐血干扰索,获得部分纯化干扰素(P-IFN)。抗病毒活性比粗制干扰素提高50倍左右,比活性为10~5单位毫克蛋白~(-1)。回收率约为37%,在纯化过程中损失较多。为了减少损失,除在操作过程中注意保冷外,并采取勤换透析液、缩短透析时间,用纤维素微孔滤膜代替蔡氏滤器,作了一些小的改进。经上进方法处理的P-IFN,效价无明显影响。用低浓度(1~3×10~4单位毫升~(-1))的P-IFN,局部注射球结膜下或浅眶内,治疗疱疹性角膜炎28例,治愈好转率为92.9%,尤以上皮型疗效最佳,治愈率为100%。此种方法具有疗效高、反应小、干扰素用量少等优点,有利于推广。
