目的观察雷公藤甲素对胶原诱导的关节炎(CIA)模型大鼠膝关节腔液肿瘤坏死因子(TNF)-琢、白细胞介素(IL)-10含量和对滑膜组织核转录因子资B(NF-资B)表达与活性的影响,探讨雷公藤治疗类风湿关节炎(RA)的可能作用机制。方法于Wistar大鼠背部及尾根部多点皮内注射牛Ⅱ型胶原乳剂制作CIA动物模型,经过雷公藤甲素治疗后,通过酶联免疫吸附(ELISA)法检测大鼠膝关节腔液TNF-琢和IL-10含量,免疫组织化学染色(SP)检测NF-资B的表达和凝胶电泳迁移分析法(electrophoreticmobilityshiftassay,EMSA)测定NF-资B活性。结果与正常对照组大鼠相比,CIA模型大鼠关节腔液TNF-琢含量升高(P<0.05),IL-10含量降低(P<0.05),膝关节滑膜组织NF-资B的表达和活性增加(P0.05)。结论雷公藤甲素可能通过下调滑膜细胞NF-资B的表达与活性,降低炎性因子的表达,升高抑炎因子的表达而治疗RA。
Objective To observe the effect of Triptolide on the content of tumor necrosis factor (TNF) - interleukin (IL) -10 in knee joint cavity fluid and the expression of NF-κB in synovial tissue of collagen-induced arthritis (CIA) (NF-B) expression and activity, to explore the possible mechanism of Tripterygium treatment of rheumatoid arthritis (RA). Methods CIA animal models were made by intradermal injection of bovine type Ⅱ collagen in the back and tail of Wistar rats. After treatment with triptolide, TNF-α in the knee joint fluid was detected by enzyme-linked immunosorbent assay (ELISA) The content of IL-10, the content of NF-kappaB, the expression of NF-kappaB and the activity of NF-kappaB were detected by electrophoretic mobility shift assay (EMSA) by immunohistochemical staining (SP) Results Compared with the normal control group, the content of TNF-α in articular cavity fluid of CIA model rats increased (P <0.05) and the content of IL-10 decreased (P <0.05), the synovial tissue NF- (P <0.05). The above indexes returned to normal after triptolide treatment (P> 0.05). Conclusion Triptolide may treat RA by down-regulating the expression and activity of NF-κB in synovial cells, decreasing the expression of inflammatory cytokines and increasing the expression of anti-inflammatory cytokines.
