目的:探讨2,4-二羟基二苯甲酮(2,4-dihydroxybenzophenone,BP-1)对可卡因致小鼠脑中多巴胺、5-羟色胺、谷氨酸和γ-氨基丁酸含量变化的影响。方法:雄性ICR小鼠随机分成1%吐温组、1%吐温+可卡因组、3个BP-1+可卡因组和3个BP-1组,采用高效液相色谱法测定并比较不同处理组小鼠脑中神经递质含量。结果:急性注射可卡因后,小鼠脑中多巴胺、5-羟色胺和γ-氨基丁酸水平显著增高;与1%吐温+可卡因组相比,BP-1(100和200 mg·kg-1)+可卡因组多巴胺水平降低,5-羟色胺含量升高;BP-1+可卡因组谷氨酸和γ-氨基丁酸含量与1%吐温+可卡因组间差异无统计学意义。结论:中、低剂量BP-1可逆转可卡因所致小鼠脑中多巴胺含量的变化,对5-羟色胺含量变化有一定影响。BP-1预给药对谷氨酸和γ-氨基丁酸含量的变化未见明显作用。
Objective: To investigate the effects of 2,4-dihydroxybenzophenone (BP-1) on the changes of dopamine, serotonin, glutamate and γ-aminobutyric acid in cocaine-induced mice brain . Methods: Male ICR mice were randomly divided into 1% Tween group, 1% Tween + cocaine group, 3 BP-1 + cocaine groups and 3 BP-1 groups. Neurotransmitter content in mouse brain. Results: Compared with 1% Tween + cocaine group, the levels of dopamine, serotonin and γ-aminobutyric acid were significantly increased in the brains of mice after acute injection of cocaine. BP-1 (100 and 200 mg · kg- + Cocaine group decreased dopamine levels, serotonin increased; BP-1 + cocaine group glutamate and γ-aminobutyric acid content and 1% Tween + cocaine group no significant difference between groups. CONCLUSION: Moderate and low doses of BP-1 can reverse the change of dopamine content in cocaine-induced mice brain, and have some influence on the content of 5-HT. BP-1 pre-administration of glutamate and γ-aminobutyric acid content had no significant effect.
