目的了解静脉吸毒HIV-1感染者Th17和Treg细胞百分比及Th17/Treg比例平衡状态,分析其与疾病进展和单核细胞免疫活化的相关性,探讨Th17/Treg比例失衡在HIV致病机制中的作用。方法选取经静脉吸毒感染HIV-1的患者58例,采集静脉血,分离外周血单个核细胞(PBMC),流式细胞术检测Th17和Treg百分比及Th17/Treg比例。ELISA方法检测血浆中sCD14水平。结果 HIV-1感染者Th17细胞百分比和Th17/Treg比值明显低于健康对照组,差异均有统计学意义(P均<0.01),Th17百分比和Th17/Treg比值与CD4数量呈正相关(P<0.01,r=0.46;P<0.01,r=0.47),与病毒载量呈负相关(P<0.01,r=-0.65;P<0.01,r=-0.69)。HIV-1感染者血浆sCD14水平显著高于健康对照组组(P<0.01),且Th17百分比和Th17/Treg比值与血浆sCD14水平呈负相关(P<0.01,r=-0.54;P<0.01,r=-0.49)。结论 HIV-1感染吸毒人群中Th17和Treg百分比和Th17/Treg比值均明显失衡,且与病毒载量和单核细胞活化标志sCD14均呈负相关,提示Th17/Treg失衡与HIV感染所致病毒复制和免疫活化密切相关。
Objective To understand the percentage of Th17 and Treg cells and the ratio of Th17 / Treg in HIV-1 infected patients, and analyze their correlation with disease progression and monocyte immune activation. To investigate the imbalance of Th17 / Treg in HIV pathogenesis effect. Methods Fifty-eight HIV infected HIV-1 patients were enrolled in this study. Venous blood was collected and peripheral blood mononuclear cells (PBMC) were isolated. The percentage of Th17 and Treg and the ratio of Th17 / Treg were determined by flow cytometry. ELISA method for detecting plasma sCD14 levels. Results The percentage of Th17 cells and the ratio of Th17 / Treg in HIV-1 infected patients were significantly lower than those in healthy controls (P <0.01). Th17 percentage and Th17 / Treg were positively correlated with CD4 count (P <0.01) , r = 0.46; P <0.01, r = 0.47), and negatively correlated with viral load (P <0.01, r = -0.65; P <0.01, r = -0.69). The level of plasma sCD14 in HIV-1 infected patients was significantly higher than that in healthy controls (P <0.01), and the Th17 percentage and Th17 / Treg were negatively correlated with plasma sCD14 (P <0.01, r = -0.54; r = -0.49). Conclusions The Th17 and Treg percentage and the Th17 / Treg ratio in HIV-1 infected population are obviously unbalanced, and negatively correlated with the viral load and monocyte activation marker sCD14, suggesting that Th17 / Treg imbalance and viral replication due to HIV infection And immune activation is closely related.
