目的探讨铅暴露人神经胶质瘤U251细胞对钙离子信号通路的影响。方法将体外培养的人神经胶质瘤U251细胞(human U251 glioma cells,U251)分别暴露于含有乙酸铅(400μg/ml)的培养液中8和24 h,提取RNA。应用cDNA芯片技术筛选差异表达基因,并利用信息学技术进行钙离子信号通路分析。结果铅暴露U251细胞,导致2840条基因差异表达,钙离子信号通路中有45条基因表达异常。结论铅暴露U251细胞后,参与钙离子信号通路的45条基因发生了表达改变,导致细胞膜上钙离子通道发生变化,钙离子调控的生物学功能受到影响。
Objective To investigate the effect of lead exposed human glioma U251 cells on calcium signaling pathway. Methods Human U251 glioma cells (U251) cultured in vitro were exposed to culture medium containing lead acetate (400 μg / ml) for 8 and 24 h, respectively. The cDNA microarray was used to screen differentially expressed genes and the information technology was used to analyze the calcium signaling pathway. Results Lead exposure to U251 cells resulted in the differential expression of 2840 genes, and 45 genes were abnormal in the calcium signaling pathway. Conclusion The lead-exposed U251 cells changed the expression of 45 genes involved in calcium signaling pathway, resulting in changes of calcium ion channels in the cell membrane and the biological function of calcium regulation.
