以含Bcl-2的重组逆转录病毒转染包装细胞系PA317,经G418抗性筛选,获得单克隆细胞,扩增并收集培养上清感染NIH3T3细胞,进而完成Bcl-2基因向NIH3T3细胞的导入。后者软琼脂集落形成率与细胞增殖速率均高于对照组,而辐射敏感性低于对照组。表明Bcl-2基因对NIH3T3细胞的增殖和辐射敏感性均具有一定的影响。
The packaging cell line PA317 was transfected with the recombinant retrovirus containing Bcl-2, and then cloned by G418 to obtain monoclonal cells. The NIH3T3 cells were expanded and collected, and the Bcl-2 gene was transduced into NIH3T3 cells . The latter soft agar colony formation rate and cell proliferation rate were higher than the control group, while the radiation sensitivity was lower than the control group. The results showed that Bcl-2 gene had some effects on the proliferation and radiosensitivity of NIH3T3 cells.
