目的:探讨细胞凋亡在局灶性脑缺血再灌流后的动态变化及其作用。方法:采用末端标记法(TUNEL)观察大鼠局灶性脑缺血再灌流不同时间在皮层区和基底节区的凋亡细胞的变化。结果:大脑中动脉阻塞2hr后随再灌流时间延长其解剖分布不同。细胞凋亡在基底节区出现时间早,持续时间短;而皮层区出现时间较晚,持续时间较长,其中再灌流1d细胞凋亡最显著。结论:本研究提示细胞凋亡与细胞坏死在局灶性脑缺血再灌流损伤中同时存在,细胞凋亡是梗塞灶形成的重要机制之一
Objective: To investigate the dynamic changes of apoptosis after focal cerebral ischemia-reperfusion and its effects. Methods: The changes of apoptotic cells in cortex and basal ganglia at different time points after focal cerebral ischemia-reperfusion in rats were observed by TUNEL. Results: After 2 hours occlusion of the middle cerebral artery, the anatomic distribution was different with the time of reperfusion. Apoptosis in the basal ganglia appeared early, short duration; and cortical areas appear later, longer duration, of which 1d after reperfusion perfusion most significant apoptosis. Conclusion: This study suggests that both apoptosis and necrosis exist simultaneously in focal cerebral ischemia-reperfusion injury. Apoptosis is an important mechanism of infarction formation
