目的 从单味中药大黄、虎杖、何首乌中提取总蒽醌类化合物 ,观察其对低密度脂蛋白氧化修饰的抑制作用。方法 以95%乙醇提取并加用碱提取酸沉淀法提取蒽醌类成分 ,观察其对 Cu2 + 诱导的 LDL氧化的抑制作用 ,以硫代巴比妥酸反应物质法(TBARS)测定丙二醛 (MDA)含量 ,计算药物对氧化的抑制率以及抑制率为 50 %时所需药物浓度 (IR5 0 )。结果 总蒽醌类提取物使氧化动力曲线迟滞相延长 ,峰值降低 ,抑制过氧化脂质生成并有浓度依赖性 ,各种药物的 IR5 0 分别为 8.1 8μg/ ml(大黄 )、1 5.58μg/ ml(虎杖 )、3.0 6× 1 0 9μg/ ml(何首乌 )。结论 三种药物蒽醌类提取物均有抑制 LDL氧化作用 ,但抑制作用强度不同。
Objective To extract the total terpenoids from single herb Rhubarb, Polygonum cuspidatum and Polygonum multiflorum, and observe its inhibitory effect on the oxidation modification of LDL. Methods Indole components were extracted with 95% ethanol and added with alkali extraction acid precipitation to observe its inhibitory effect on Cu2 + induced LDL oxidation. Malondialdehyde was measured by thiobarbituric acid reactive substance method (TBARS). (MDA) content, calculated drug inhibition rate of oxidation, and the required drug concentration (IR50) when the inhibition rate was 50%. Results The total anthraquinone extracts prolonged the delayed oxidation kinetic curve, reduced the peak value, and inhibited the production of peroxide lipids in a concentration-dependent manner. The IR50 values of various drugs were 8.18 μg/ml (rhubarb) and 15.58 μg/, respectively. Ml (Polygonum cuspidatum), 3.0 6 × 10 9 μg/ml (Polygonum multiflorum). Conclusion All three kinds of drug steroid extracts inhibit LDL oxidation, but their inhibitory effects are different.
