以刺芹侧耳不同生长发育期的基质菌丝体和子实体为试材,采用氨基酸自动分析仪对刺芹侧耳栽培过程中基质菌丝体和子实体的游离氨基酸含量的动态变化进行了研究,以期掌握刺芹侧耳整个栽培周期中基质菌丝体和子实体中氨基酸含量的变化动态,为进一步探索刺芹侧耳栽培过程中菌丝吸收利用氮后合成的有机氮载体以及转运和传递方式奠定基础。结果表明:基质菌丝体中总游离氨基酸含量在发菌阶段显著上升,在后熟阶段及搔菌处理时期,总游离氨基酸含量变化不显著;子实体生长阶段,基质菌丝体中总游离氨基酸含量逐渐下降。基质菌丝体中含量最丰富的氨基酸为精氨酸(Arg),现原基时含量达到最高值为7 082.30μg·g~(-1)(占比65.86%),从现原基到子实体成熟过程中基质菌丝体中Arg含量逐渐下降,最终为1 741.31μg·g~(-1)(占比40.67%)。由此推测Arg在刺芹侧耳生长过程中的氮素营养转运中起到重要作用。
Amino acid automatic analyzer was used to study the dynamic changes of free amino acid content in the mycelium and fruiting bodies of Pleurotus tuberosus during the cultivation of Pleurotus tuberosus L. with different growth and development stages. Aristolochia Auriculae during the entire cultivation period of the matrix mycelium and fruiting bodies of amino acid content changes dynamic, to further explore the Ceratoides argyi cultivation mycelium absorption and utilization of nitrogen after the synthesis of organic nitrogen carrier and lay the foundation for transport and delivery. The results showed that the content of total free amino acids in the mycelium of the substrate increased significantly in the stage of fungus, and the content of total free amino acids did not change significantly in the stage of ripening and the stage of scratching bacteria. In the fruiting body growth phase, The content gradually decreased. Arginine (Arg) is the most abundant amino acid in the mycelia of the matrix, and the highest content of the original mycelium is 7 082.30 μg · g -1 (accounting for 65.86%), Arg content in the matrix mycelium gradually decreased during solid maturation, eventually reaching 1 741.31 μg · g -1 (accounting for 40.67%). This suggests that Arg plays an important role in the nitrogen nutrition transport during the growth of Pleurotus citrinopileatus.
