目的建立兔血浆中吴茱萸碱和吴茱萸次碱浓度的测定方法,测定吴茱萸碱和吴茱萸次碱的血浆蛋白结合率。方法采用平衡透析法测定吴茱萸碱和吴茱萸次碱血浆蛋白结合率,利用HPLC测定血浆中吴茱萸碱和吴茱萸次碱浓度。结果吴茱萸碱在0.208,1.04,2.08μg.mL-1浓度下的血浆蛋白结合率分别为(38.5±1.7)%、(36.4±1.0)%、(39.0±1.0)%,吴茱萸次碱在0.235,1.18,2.345μg.mL-1浓度下的血浆蛋白结合率分别为(40.6±3.2)%、(39.3±4.5)%、(41.5±1.2)%,吴茱萸碱和吴茱萸次碱的浓度对血浆蛋白结合率无显著影响。结论吴茱萸碱和吴茱萸次碱与兔血浆蛋白具有较低等强度的结合。
Objective To establish a method for the determination of evodiamine and rutaecarpine in rabbit plasma and determine the plasma protein binding rate of evodiamine and rutaecarpine. Methods The plasma protein binding rate of evodiamine and rutaecarpine was determined by equilibrium dialysis. The concentrations of evodiamine and rutaecarpine in plasma were determined by HPLC. Results The plasma protein binding rates of evodiamine at the concentrations of 0.208, 1.04 and 2.08μg.mL-1 were (38.5 ± 1.7)%, (36.4 ± 1.0)%, (39.0 ± 1.0)% and rutaecarpine at 0.235, The plasma protein binding rates were (40.6 ± 3.2)%, (39.3 ± 4.5)% and (41.5 ± 1.2)% at concentrations of 1.18 and 2.345μg.mL-1 respectively. The concentrations of evodiamine and rutaecarpine in plasma protein binding No significant impact on rates. Conclusions Evodiamine and rutaecarpine have a lower intensity of binding to rabbit plasma proteins.
