目的:研究生物波调控因子对脑缺血后再灌流脑细胞线粒体有无保护作用。方法:通过大鼠四血管闭塞(热灼两侧椎动脉、夹闭两侧颈总动脉)造成脑缺血和再灌流的动物模型,提纯脑组织匀浆中线粒体并用丙酮酸钠及苹果酸钠作底物,测定脑细胞线粒体的呼吸三态、四态和呼吸控制率。结果:脑缺血20min组和缺血20min后再灌流4d组大鼠脑线粒体呼吸功能与假手术组相比差异非常显著,特别是呼吸控制率的降低非常明显;而用生物波调控因子治疗的缺血20min再灌流4d的大鼠无论呼吸三态、四态及呼吸控制率都与假手术组差异不明显。结论:生物波调控因子对缺血后再灌流性脑损伤的脑线粒体呼吸功能有一定保护作用,研究可能为治疗缺血性脑损伤开辟一条新途径。
Objective: To study the protective effects of bio-wave regulators on mitochondria of cerebral cells after cerebral ischemia and reperfusion. Methods: Animal models of cerebral ischemia and reperfusion caused by occlusion of the four vessels in rats (thermal cauterization of both vertebral arteries and occlusion of common carotid arteries on both sides) were performed. The mitochondria in brain homogenates were purified and treated with sodium pyruvate and sodium malate As substrate, measuring the mitochondria of respiratory cells of the three-state, four-state and respiratory control. Results: There were significant differences in mitochondrial respiratory function between the 20 min ischemia group and the 4 d ischemia reperfusion group and the sham operation group, especially the decrease of respiration control rate. However, No significant difference was found between the four states and the rate of respiration control in sham-operation group. Conclusion: The bio-wave regulators play a protective role in mitochondrial respiratory function after cerebral ischemia-reperfusion injury, and the study may open up a new way for the treatment of ischemic brain injury.
