目的研究当归和川芎对尾吊大鼠比目鱼肌(SOL)肌球蛋白重链(MHC)表达及肌萎缩的影响。方法尾部悬吊法建立模拟失重模型,将大鼠分为同步饲养组(Con)、尾吊灌溶媒组(HLU+W)、尾吊当归灌胃组(HLU+Ang)和尾吊川芎灌胃组(HLU+Lig)。免疫组化法检测大鼠比目鱼肌肌球蛋白重链的表达。结果与同步饲养组相比,尾吊组比目鱼肌的湿重,肌重体重比以及I型和II型肌纤维横截面积均有所下降,而MHCII的表达和II型肌纤维的比例上升。与未治疗组相比,当归可以使I型和II型肌纤维横截面积分别提高29%和35%,MHCII的表达下降。川芎可使I型肌纤维横截面积提高25%(P=0.064)并相应减少MHCII的表达。结论当归和川芎均能显著抑制模拟失重导致的MHCII型表达的升高,并缓解尾吊大鼠肌纤维横截面积的缩小。
Objective To study the effects of Angelica sinensis and Chuanxiong on the expression of myosin heavy chain (MHC) and muscle atrophy in soleus muscle (SOL) of tail-suspended rats. Methods The model of weightlessness was established by tail suspension method. The rats were divided into three groups: Con group, HLU + W group, HLU + Ang group and Ligusticum chuanxiong group Group (HLU + Lig). Immunohistochemistry was used to detect the expression of myosin heavy chain in rat soleus. Results Compared with the synchronized feeding group, the wet weight, the body weight ratio and the cross-sectional area of type I and type II myofibers in the tail suspension group decreased, while the expression of MHC II and the proportion of type II myofibers increased. Compared with untreated group, Angelica sinensis could increase the cross-sectional area of type I and type II myofibers by 29% and 35%, respectively, and the expression of MHCII decreased. Chuanxiong could increase the cross-sectional area of type I muscle fiber by 25% (P = 0.064) and reduce the expression of MHCII. Conclusion Both Angelica sinensis and Chuanxiong rhizome could significantly inhibit the increase of MHC II expression induced by simulated weightlessness and alleviate the narrowing of muscle fiber cross-sectional area in tail-suspended rats.
