目的观察芪归解毒汤药物血清对硫酸镍损伤正常人晶状体上皮细胞(Lens epithium cells,LEC,细胞编号:SRA01/04)的保护作用。方法采用血清药理学方法提取芪归解毒汤药物血清,体外培养晶状体上皮细胞,用硫酸镍产生细胞损伤,再加入体积分数分别为5%、10%、15%、20%的药物血清,倒置显微镜下观察晶状体上皮细胞的形态学改变,MTT法检测细胞活性的变化,试剂盒测量乳酸脱氢酶(LDH)含量的变化。结果倒置显微镜下观察到硫酸镍损伤的晶状体上皮细胞出现肿胀、体积缩小、折光性下降、贴壁率降低等形态学改变。加入10%和15%芪归解毒汤药物血清时细胞生长明显好转,折光性优于硫酸镍组,药物血清浓度为20%时细胞生长接近正常;MTT法检测药物血清组细胞活力较硫酸镍组明显增强;药物血清组LDH活性较硫酸镍组明显降低。结论芪归解毒汤药物血清能明显减轻晶状体上皮细胞的镍性损伤,并呈现剂量依赖关系。
Objective To observe the protective effect of the serum of Zenggui Jiedu Decoction on the damage of normal human lens epithelial cells (LEC, cell number: SRA01/04) by nickel sulfate. Methods Serum of pharmacological method was used to extract the serum of Zonggui Jiedu Decoction. Lens epithelial cells were cultured in vitro. Cell injury was induced by nickel sulfate. Then serum with drug concentration of 5%, 10%, 15%, and 20% was added. Inverted microscope The morphological changes of the lens epithelial cells were observed, the cell viability was measured by the MTT assay, and the kit was used to measure changes in the lactate dehydrogenase (LDH) content. RESULTS: Under the inverted microscope, the lens epithelial cells damaged by nickel sulphate were observed to have morphological changes such as swelling, reduced volume, decreased refractive index, and decreased adherence rate. Adding 10% and 15% Zigui Jiedu Decoction drug serum significantly improved cell growth, refraction better than the nickel sulfate group, the drug serum concentration of 20% when the cell growth was close to normal; MTT assay drug serum group cell viability compared to nickel sulfate group Significantly increased; LDH activity in the drug serum group was significantly lower than that of the nickel sulfate group. Conclusion The serum of Zenggui Jiedu Decoction can significantly reduce the nickel damage of lens epithelial cells and show a dose-dependent relationship.
