目的:观察脑出血大鼠心肌组织HSP70蛋白表达的变化及扶正护脑胶囊的干预作用。方法:动物随机分为假手术组、模型组、扶正护脑组和安宫牛黄组,分别于术后6h、24h、72h 3个时相点取材,采用免疫组织化学SABC法测定心肌组织HSP70蛋白表达,并进行图像分析,观测其IOD值;并与血清CK-MB、cTnI含量作相关性分析。结果:HSP70阳性颗粒主要在心内膜下的心肌细胞和血管内皮细胞的胞浆中表达。模型组各时间点心肌组织HSP70蛋白表达均显著升高,其IOD值亦明显升高,扶正护脑组和安宫牛黄组均能明显增加HSP70蛋白的表达,升高其IOD值。IOD值与cTnI、CK-MB含量呈正相关。结论:HSP70对脑出血诱发的心肌损伤具有保护作用;扶正护脑胶囊通过提高HSP70的水平,减轻脑出血后心肌的损伤。
Objective: To observe the changes of HSP70 protein expression in rats with intracerebral hemorrhage and the intervention effect of Fuzheng protects brain capsule. METHODS: Animals were randomly divided into sham-operated group, model group, Fuzhenghong group and Angong Niuhuang group. The rats were obtained at three time points (6h, 24h, and 72h after operation). Myocardial HSP70 protein was determined by immunohistochemical SABC method. Expression, and image analysis, observation of the IOD value; and serum CK-MB, cTnI content for correlation analysis. RESULTS: HSP70 positive particles were mainly expressed in the cytoplasm of subendocardial myocytes and vascular endothelial cells. The expression of HSP70 protein in the myocardium of the model group was significantly increased at each time point, and the IOD value was also significantly increased. Both the Fuzhenghong group and the Angong Niuhuang group could significantly increase the expression of HSP70 protein and increase the IOD value. IOD values were positively correlated with cTnI and CK-MB levels. Conclusion: HSP70 has a protective effect on myocardial injury induced by cerebral hemorrhage; Fuzheng Huonao capsule can reduce the damage of myocardium after cerebral hemorrhage by increasing the level of HSP70.
