为了预防由2型单纯疱疹病毒(HSV-2) 引起的原发和复发感染,作者利用基因工程方法构建了糖蛋白H(gH)基因缺失的HSV-2病毒株和表达gH的Vero细胞系.由此细胞系产生的病毒能感染正常细胞,完成一个复制周期,但由其他细胞产生的病毒则因缺乏gH基因而没有感染性,故称为不能感染的单周期(DISC)病毒.作者以豚鼠为对象评价了HSV-2 DISC的效果.疫苗接种方式为经阴道(ivag)和皮下注射(sc)两种.采用双盲法,根据疱疹原发和复发阶段外阴损害数目和红斑的严重程度进行临床计分以判断症状的严重程度.在Vero细胞中测定蚀斑,以中和试验和ELISA测血清抗体滴度.
To prevent primary and recurrent infections caused by herpes simplex virus type 2 (HSV-2), the authors constructed a strain of HSV-2 with a deletion of the glycoprotein H (gH) gene and a Vero cell line expressing gH using genetic engineering. The virus produced by this cell line can infect normal cells and complete a replication cycle, but the viruses produced by other cells are not infectious due to the lack of gH gene, so it is called unicerable single-cycle (DISC) virus. The effect of HSV-2 DISC was evaluated for both vaccination with ivag and subcutaneous (sc) vaccination using a double-blind method based on the number of vulvar lesions and the severity of erythema in the primary and recurrent stages of herpes Clinical scores were used to determine the severity of the symptoms. Plaque was measured in Vero cells and serum antibody titers were determined by neutralization and ELISA.
