为探讨新型抗氧化剂SS31在体外对细胞的辐射防护作用和初步机制,用60Coγ射线照射细胞,用不同浓度的SS31对细胞进行干预,观察照射后细胞的增殖、生存状态、细胞内ROS水平和线粒体膜电位的改变,并将药物干预组和阴性对照组进行比较。结果发现:7.5 Gy的γ射线照射后24–72 h,细胞增殖活性显著降低;48 h内细胞凋亡和坏死比率显著增加,并伴随胞内ROS增长、线粒体膜电位下降和细胞周期阻滞。SS31能明显改善这些反应,药物干预组与阴性对照相比有统计学差异。通过本实验,观察到了SS31具有明显的体外辐射防护作用,提示其有潜力作为一种新型的辐射防护剂。
In order to explore the protective effect and preliminary mechanism of the new antioxidant SS31 on cells in vitro, the cells were irradiated with 60Coγ ray and the cells were treated with different concentrations of SS31 to observe the proliferation, survival status, intracellular ROS levels and mitochondria Membrane potential changes, and the drug intervention group and negative control group were compared. The results showed that: after 24-72 h irradiation with 7.5 Gy γ-rays, the cell proliferation activity was significantly reduced; the apoptosis and necrosis rates were significantly increased within 48 h, accompanied by the intracellular ROS growth, decreased mitochondrial membrane potential and cell cycle arrest. SS31 can significantly improve these responses, the drug intervention group compared with the negative control was statistically significant. Through this experiment, it was observed that SS31 has obvious radiation protective effect in vitro, suggesting that it has the potential as a new type of radioprotectant.
