目的:探讨nm23-H1基因表达与大肠癌转移的关系。方法:应用过氧化酶标记的链霉卵白素(SP)染色法对33例正常大肠粘膜、134例癌旁粘膜及193例大肠癌组织进行免疫组织化学染色,并对染色结果进行观察。结果:大肠粘膜与癌旁粘膜间nm23-H1表达阳性率差异无显著性(P>0.05),但nm23-H1在良、恶性大肠癌组织中的表达阳性率差异有显著性(P<0.05~0.01)。nm23-H1表达阳性率仅在正常、良性大肠组织与转移的大肠癌间差异有显著性(P<0.01),但与未转移大肠癌间差异无显著性(P>0.05)。在已发生转移的大肠癌组织中,nm23-H1表达阳性率和强度明显低于尚未发生转移者(P<0.01)。在39例无淋巴结转移的大肠癌组织中未见nm23-H1表达,而在24例转移的大肠癌中可检测到nm23-H1表达。结论:nm23-H1在抑制大肠癌转移中发挥一定作用,但在大肠癌转移过程中可能还有其他调节途径/因素存在。
Objective: To investigate the relationship between nm23-H1 gene expression and metastasis of colorectal cancer. METHODS: Immunohistochemical staining of 33 cases of normal colorectal mucosa, 134 cases of paraneoplastic mucosa and 193 cases of colorectal cancer tissues were performed using peroxidase-labeled streptavidin (SP) staining method. The staining results were observed. Results: There was no significant difference in the expression of nm23-H1 between colorectal mucosa and paratumor mucosa (P>0.05). However, the positive expression rate of nm23-H1 in benign and malignant colorectal cancer tissues was significantly different (P< 0.05 to 0.01). The positive rate of nm23-H1 expression was only significant between normal and benign colorectal tissues and metastatic colorectal cancer (P<0.01), but there was no significant difference between normal and benign large intestine tissue and metastatic colorectal cancer (P>0.05). In metastatic colorectal cancer tissues, the positive rate and intensity of nm23-H1 expression were significantly lower than those without metastasis (P<0.01). There was no expression of nm23-H1 in 39 colorectal cancer tissues without lymph node metastasis, but nm23-H1 expression was detected in 24 metastatic colorectal cancers. Conclusion: nm23-H1 plays a role in the inhibition of metastasis of colorectal cancer, but there may be other regulatory pathways/factors in the metastasis of colorectal cancer.
