目的观察青刺果多糖对四氧嘧啶诱导的糖尿病小鼠心肌组织病变的影响。方法腹腔一次性注射四氧嘧啶复制模型。造模成功后,将实验动物分为模型组和青刺果多糖治疗组(300mg/kg),另设正常组。实验小鼠分别于给药2周末和4周末测定空腹12h血糖,然后随机处死,取心固定于4%多聚甲醛溶液中,制作病理切片,经苏木素-伊红(HE)染色后光镜下观察。结果青刺果多糖治疗组与模型组相比,血糖显著降低,差异有统计学意义(P<0.05);光镜下,模型组小鼠心肌纤维纵横纹模糊,心肌细胞有颗粒变性、脂肪变性,部分区域心肌纤维互相融合,可见呈灶性分布的嗜酸性变的心肌纤维,并伴有坏死、纵横纹消失、间质内纤维母细胞增生,严重者可见心肌纤维化形成的瘢痕。经青刺果多糖治疗后,心肌纤维纵横纹清晰,排列整齐,呈性分布的嗜酸性变的心肌纤维减少,可见少量的纤维细胞增生。结论青刺果多糖对糖尿病小鼠心肌具有保护作用。
Objective To observe the effect of polysaccharides of prickly fruit on myocardial pathological changes in alloxan-induced diabetic mice. Methods A one-time intraperitoneal injection of alloxan replication model. After successful modeling, the experimental animals were divided into model group and prickly fruit polysaccharide treatment group (300mg/kg), and the normal group was set up. Measure fasting 12h blood glucose in experimental mice on the 2nd and 4th week after administration, and then randomly sacrificed, and coring fixed in 4% paraformaldehyde solution to make pathological sections, stained with hematoxylin-eosin (HE) under light microscopy. Observed. Results Compared with the model group, the blood glucose of the prickly pear fruit polysaccharide treatment group was significantly reduced (P<0.05). Under the light microscope, the cross-stained pattern of myocardial fibers was obscure in the model group, and the myocardial cells had degeneration and steatosis. Some areas of myocardial fiber fusion with each other, showing a focal distribution of eosinophilic myocardium fibers, accompanied by necrosis, cross-sectional pattern disappeared, interstitial fibroblast proliferation, severe myocardial fibrosis can be seen in the formation of scars. After treatment with P. sinensis polysaccharides, the cross-sectional pattern of myocardial fibers was clear, neatly arranged, and the eosinophilic myocardial fibers that were distributed were decreased, showing a small amount of fibrocyte proliferation. Conclusion The polysaccharide of P. truncatum has a protective effect on the myocardium of diabetic mice.
