目的 为了选择贝母乙素含量较高的贝母原材料。方法 建立了 HPL C法分析贝母原材料中贝母乙素含量。贝母生药粉用氨水 - 95 %乙醇 (1∶ 5 )混合液提取 ,柱前用 2 ,4-二硝基苯肼 (DNPH)衍生 ,衍生化条件 5 0℃下反应 30 min。分析色谱柱 :Hypersil C1 8ODS2 ,流动相 :CH3CN- 10 0 mmol/ L CH3COONH4(4 1∶ 5 9,p H=5 .0 ) ,流速为1.2 m L/ min。U V375 nm检测。结果 4种市售贝母中东贝母的贝母乙素含量最高。结论 方法灵敏 ,重现性好 ,样品用量少 ,便于应用
Objective To select high Fritillariae Fritillaria raw materials. Methods The HPL C method was established to analyze the content of ferritin in Fritillaria raw materials. Fritillaria powder was extracted with a mixture of ammonia-95% ethanol (1:5) and derivatized with 2,4-dinitrophenylhydrazine (DNPH) before derivatization. The reaction conditions were derivatized at 50 °C for 30 min. Analysis column: Hypersil C1 8ODS2, mobile phase: CH3CN- 10 0 mmol/L CH3COONH4 (4 1: 5 9, p H = 5.0), flow rate 1.2 m L/min. U V375 nm detection. RESULTS Four species of Fritillaria meleagris in the market were found to have the highest content of pemetrexine. Conclusion The method is sensitive, reproducible, and the amount of sample is small for easy application
