目的 研究高温对整体大鼠腹腔巨噬细胞吞噬功能及凋亡的影响。方法 Wistar大鼠在 (40± 0 .2 )℃作用下 ,维持肛温 40℃ 1h ,恢复室温后 ,用透射电子显微镜观察在刀豆素A刺激下 ,分别在 5、15、30、6 0、12 0min巨噬细胞受体介导功能的改变以及凋亡的出现。结果 腹腔注射刀豆素A后 ,5min时即观察到含大量金颗粒的内吞体、溶酶体增多 ,且一直持续 ,15min时金颗粒和溶酶体出现融合 ,但 6 0、12 0min尚可观察到未完全排放的金颗粒 ;同时观察到巨噬细胞核固缩以及自噬体增多。结论 巨噬细胞内吞增强 ,酸性磷酸酶活力增高 ,但是 ,外排能力减弱 ,并出现凋亡。
Objective To study the effects of high temperature on phagocytosis and apoptosis of rat peritoneal macrophages. Methods The Wistar rats were maintained at 40°C for 1 h at (40 ± 0.2)°C. After recovering at room temperature, they were observed under the stimulation of Concanavalin A at 5, 15, 30, and 60, respectively. , 120 min change in macrophage receptor-mediated function and appearance of apoptosis. RESULTS After intraperitoneal injection of concanavalin A, the endosomes and lysosomes containing large amounts of gold particles were observed at 5 min and continued to persist. At 15 min, gold particles and lysosomes appeared to fuse, but 60 min and 120 min remained. Incomplete discharge of gold particles can be observed; macrophages nuclear condensation and increased autophagosomes are also observed. Conclusions The macrophage endocytosis is enhanced and the acid phosphatase activity is increased. However, the efflux ability is decreased and apoptosis occurs.
