目的考察从含阿魏酸的小鼠血浆中提取分离微量阿魏酸并制成HPLC微量检测样品的不同方法。方法在小鼠血浆中外加一定量的阿魏酸,分别用甲醇法、乙醇法、加热法提取分离小鼠血浆中微量阿魏酸并进行HPLC检测。结果用乙醇法提取分离小鼠血浆中微量阿魏酸的方法最适用于HPLC检测。结论乙醇法简便快速,重现性好,稳定性强,提取分离效果好,是较理想的小鼠血浆中微量阿魏酸提取分离方法;适用于从小鼠血浆中提取分离微量阿魏酸并用于HPLC法测定小鼠体内阿魏酸血药浓度及药代动力学研究。
Objective To investigate the different methods of extracting trace ferulic acid from plasma of ferulic acid-containing mice and making HPLC micro-assay. Methods A certain amount of ferulic acid was added to the plasma of mice. The trace amounts of ferulic acid in the plasma of mice were extracted and isolated by methanol, ethanol, and heating methods and detected by HPLC. Results The method of extracting trace ferulic acid from mouse plasma by ethanol extraction method was most suitable for HPLC detection. Conclusion The ethanol method is simple, rapid, reproducible, stable, and has good extraction and separation effect. It is an ideal extraction and isolation method for trace ferulic acid in mouse plasma. It is suitable for the extraction and isolation of trace ferulic acid from mouse plasma and is used in Determination of ferulic acid plasma concentration and pharmacokinetics in mice by HPLC.
