目的探讨苦参碱(Ma)对体外培养的人视网膜母细胞瘤HXO-Rb44细胞的生长抑制作用及其机制。方法培养的人HXO-Rb44细胞经不同浓度的Ma作用后。采用噻唑蓝比色法(MTT法)测定细胞的生长状况;采用苏木素-伊红染色、流式细胞技术及核苷酸末端转移酶介导的dUTP缺口标记法检测细胞凋亡率和形态学变化;采用端粒重复扩增法检测端粒酶活性的变化。结果一定浓度范围内Ma可抑制HXO-Rb44细胞的生长(P<0.01),24 h内呈浓度依赖性。0.4 mmol/L Ma作用12 h,可观察到HXO-Rb44细胞凋亡的形态改变,细胞凋亡率开始增加(P<0.01),端粒酶活性下降(P <0.01),两者在48 h内均呈时间依赖性。并具有一定相关性(r=-0.961,P<0.01)。结论Ma可抑制HXO-Rb44细胞生长,诱导其凋亡,端粒酶活性下降可能是Ma诱发HXO-Rb44细胞凋亡机制之一,提示Ma对视网膜母细胞瘤的综合治疗具有一定的应用前景。
Objective To investigate the inhibitory effect of matrine (Ma) on the growth of human retinoblastoma HXO-Rb44 cells and its mechanism. Methods Human HXO-Rb44 cells cultured with different concentrations of Ma after action. The cell growth was measured by MTT method. The apoptotic rate and morphological changes were detected by hematoxylin-eosin staining, flow cytometry and nucleotide nick-end-transferase-mediated dUTP nick labeling The telomerase activity was detected by telomeric repeat amplification. Results Ma inhibited the growth of HXO-Rb44 cells in a certain concentration range (P <0.01), and showed a concentration-dependent manner within 24 h. The morphological changes of apoptosis of HXO-Rb44 cells were observed after treated with 0.4 mmol / L Ma for 12 h. The apoptotic rate of HXO-Rb44 cells began to increase (P <0.01) and telomerase activity decreased (P <0.01) , Both in a time-dependent manner within 48 h. And had a certain correlation (r = -0.961, P <0.01). Conclusion Ma inhibits the growth of HXO-Rb44 cells and induces apoptosis. The decrease of telomerase activity may be one of the mechanisms of Ma-induced apoptosis in HXO-Rb44 cells. This suggests that Ma may have some potential applications in the comprehensive treatment of retinoblastoma.
