目的:探讨饮酒对高脂血症大鼠口服阿托伐他汀疗效及安全性的影响。方法:采用喂养高脂饲料建立高脂血症大鼠模型,将大鼠随机分为模型组、阿托伐他汀组和饮酒组。模型组大鼠灌胃蒸馏水,阿托伐他汀组大鼠灌胃10 mg/kg阿托伐他汀,饮酒组大鼠灌胃10 mg/kg阿托伐他汀+5 ml/kg白酒,连续8周。比较三组大鼠血清总胆固醇(total cholesterol,TG)、甘油三酯(triglyceride,TC)、低密度脂蛋白胆固醇(low density lipoprotein cholestorol,LDL-C)、高密度脂蛋白胆固醇(high density lipoprotein cholestorol,HDL-C)、谷草转氨酶(aspertate transarninase,AST)、谷丙转氨酶(alanine transarninase,ALT)、肌酐(creatinine,Cr)以及尿素氮(blood urea nitrogen,BUN)水平。结果 :与阿托伐他汀组比较,饮酒组大鼠的TG水平显著升高,TC、LDL-C水平稍微升高,HDL-C水平稍微降低;AST和ALT水平显著升高;Cr和BUN水平显著升高。结论 :饮酒能够减弱阿托伐他汀的降TG效果,增加阿托伐他汀的肝毒性和肾毒性。
Objective: To investigate the effect of drinking alcohol on the efficacy and safety of oral atorvastatin in hyperlipidemic rats. Methods: Hyperlipidemia rats were established by feeding high-fat diet. The rats were randomly divided into model group, atorvastatin group and drinking group. Rats in the model group were fed with distilled water. Atorvastatin group was given 10 mg / kg atorvastatin. Rats in drinking group were given 10 mg / kg atorvastatin + 5 ml / kg liquor for 8 weeks . The levels of serum total cholesterol (TG), triglyceride (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholestorol (HDL-C), aspertate transaminase (AST), alanine transaminase (ALT), creatinine (Cr) and blood urea nitrogen (BUN) Results: Compared with the atorvastatin group, TG levels were significantly increased in the alcohol drinking group, TC, LDL-C levels were slightly increased, HDL-C levels were slightly decreased, AST and ALT levels were significantly increased, Cr and BUN levels Significantly increased. Conclusions: Drinking alcohol can attenuate the hypoglycemic effect of atorvastatin and increase the hepatotoxicity and nephrotoxicity of atorvastatin.
