目的建立一种新的深低温停循环(DHCA)脑缺血/保护大鼠模型。方法SD大鼠40只,分为实验组和对照组。麻醉后气管插管接小动物呼吸机,股动脉置管测动脉压力,监测心电、体温、血氧饱和度和血气分析,用冰块将大鼠体温降至21℃。实验组阻断双侧颈总动脉60min(对照组不阻断颈总动脉)后逐渐将体温恢复正常。结果实验组4只大鼠复苏失败死亡,16只大鼠成活,死亡率为20%,4h后Longa5级评分标准评定动物神经系统损伤为1~2级,24h后无神经系统受损表现;对照组无大鼠死亡,4h后Longa评分为0级。结论本实验方法制作的大鼠模型可以用于深低温停循环脑缺血/保护的研究。
Objective To establish a new hypothermic circulatory arrest (DHCA) cerebral ischemia / protection rat model. Methods 40 SD rats were divided into experimental group and control group. After anesthesia tracheal intubation connected to small animal ventilator, femoral artery catheter arterial pressure measurement, monitoring of ECG, body temperature, oxygen saturation and blood gas analysis, the body temperature with ice to 21 ℃. Experimental group blocked bilateral common carotid artery 60min (control group does not block the common carotid artery) and gradually returned to normal body temperature. Results In the experimental group, 4 rats died of resuscitation failure and 16 rats survived with a mortality rate of 20%. After 4 hours, the Longa 5 grade scoring system rated the neurological injury as grade 1 or 2 and no neurological damage after 24 h. The rats in the group were killed, and the Longa score was 0 after 4 hours. Conclusion The rat model made in this experiment can be used to study cerebral ischemia / protection in hypothermic circulatory arrest.
