目的探讨神经肽Y(NPY)对EAE豚鼠发病及对Th1/Th2细胞平衡的影响,探索NPY对EAE发病影响的免疫调节机制。方法 30只豚鼠随机分为正常对照组、EAE对照组、NPY干预组。观察三组豚鼠血清IL-4、IFN-γ的含量,Th1/Th2比值,以及发病情况。结果 EAE豚鼠发病后血清IL-4含量降低,血清IFN-γ含量升高,Th1/Th2比值升高,Th1/Th2细胞失衡;而NPY干预组高峰期血清IL-4含量明显增高(P<0.001),血清IFN-γ含量明显降低(P<0.001),Th1/Th2比值降低(P<0.001),一定程度地纠正Th1/Th2细胞失衡。结论侧脑室注射NPY对EAE能起到保护作用,其保护机制可能是通过调节Th1/Th2细胞平衡,促进IL-4分泌,抑制IFN-γ的产生而发挥。
Objective To investigate the effect of neuropeptide Y (NPY) on the pathogenesis of EAE in guinea pigs and the balance of Th1 / Th2 cells and to explore the immunoregulatory mechanism of NPY on the pathogenesis of EAE. Methods Thirty guinea pigs were randomly divided into normal control group, EAE control group and NPY intervention group. The levels of IL-4, IFN-γ, Th1 / Th2, and incidence of the three groups of guinea pigs were observed. Results After the onset of EAE, the levels of IL-4 in serum and IFN-γ in serum increased, the ratio of Th1 / Th2 increased and the imbalance of Th1 / Th2 cells was observed in serum of EAE guinea pigs (P <0.001) ), Serum IFN-γ levels were significantly lower (P <0.001), Th1 / Th2 ratio was lower (P <0.001), to a certain extent, to correct the imbalance of Th1 / Th2 cells. Conclusion Intraventricular injection of NPY can protect EAE. The protective mechanism may be through regulating the balance of Th1 / Th2 cells, promoting the secretion of IL-4 and inhibiting the production of IFN-γ.
