目的检测肿瘤坏死因子相关凋亡诱导配体(TRAIL)、核因子-κB(NF-κB)在胎膜早破(PROM)孕妇胎膜组织中的表达水平及临床意义。方法以2012年6月-2015年12月期间在该院分娩的80例足月胎膜早破患者为研究对象(设为研究组),并以同期80例正常妊娠妇女为对照(设为正常组),用RT-PCR法和SP法检测2组胎膜组织的TRAIL、NF-κB水平。结果研究组TRAIL、NF-κB的阳性表达率(88.75%,91.25%)均高于正常组(58.75%,61.25%),差异有统计学意义(χ~2值分别为6.527、6.998,均P<0.05);研究组中TRAIL mRNA、NF-κB mRNA水平(1.71±0.16、4.28±0.16)均高于正常组(1.01±0.13、1.07±0.15),差异有统计学意义(t值分别为2.115、1.957,均P<0.05)。研究组中TRAIL与NF-κB表达呈显著正相关关系(r=0.793,P<0.05)。结论 TRAIL、NF-κB在胎膜早破孕妇胎膜组织中存在高表达,且TRAIL与NF-κB在胎膜早破孕妇胎膜组织中的表达密切相关,显示二者在胎膜早破发病中发挥着重要作用,可作为相关临床检查诊断的参考指标。
Objective To detect the expression of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and nuclear factor-κB (NF-κB) in fetal membranes of pregnant women with premature rupture of membranes (PROM) and its clinical significance. Methods Eighty patients with full-term premature rupture of membranes in our hospital from June 2012 to December 2015 were enrolled in this study. The study group consisted of 80 normal pregnant women (normal) Group). The levels of TRAIL and NF-κB in fetal membranes were detected by RT-PCR and SP method. Results The positive rates of TRAIL and NF-κB in the study group (88.75%, 91.25%) were significantly higher than those in the normal group (58.75% and 61.25% respectively) (χ ~ 2 = 6.527 and 6.998, respectively, P <0.05). The TRAIL mRNA and NF-κB mRNA levels (1.71 ± 0.16 and 4.28 ± 0.16) in the study group were significantly higher than those in the normal group (1.01 ± 0.13 and 1.07 ± 0.15 respectively) (t = 2.115 , 1.957, all P <0.05). There was a significant positive correlation between the expression of TRAIL and NF-κB in the study group (r = 0.793, P <0.05). Conclusion The expression of TRAIL and NF-κB in fetal membranes of pregnant women with premature rupture of membranes is high, and the expression of TRAIL and NF-κB in the fetal membranes of premature rupture of membranes is closely related, indicating that TRAIL and NF- Play an important role, as a reference for the diagnosis of related clinical indicators.
