目的 了解辽宁省艾滋病病毒 1(HIV 1)感染者 艾滋病 (AIDS)患者体内不同亚型HIV 1膜蛋白V3环氨基酸序列特征 ,突变种类特点。方法 提取含有整合HIV 1前病毒的基因组DNA ,巢式聚合酶链反应 (nest PCR)扩增后直接测序 ,并做序列排列比对、翻译和分析。结果 辽宁省HIV 1感染者感染病毒分属A、B’、C、G四种亚型 ,AIDS患者体内病毒V3环发生与T嗜性 SI表型有关的氨基酸突变形式 (11位出现R ,13位出现S、T、N ,19位出现V ,2 0位出现Y ,2 5、2 9位出现N等 )高于无症状感染组 (P <0 .0 5 ) ,此外还发现GQGR、APGQ、RPGA、GLGR、RPGA等少见的V3环顶端四肽组成形式及第 5位H ,34位S、F等罕见突变形式。结论 辽宁省AIDS人群中 ,A、B、C、G亚型HIV 1毒株V3环各位置出现氨基酸突变情况与国外对B亚型毒株的研究结果总体上相似 ,但发现一些罕见突变和少见V3环顶端四肽组成形式。
Objective To investigate the amino acid sequence of HIV-1 membrane proteins of different subtypes in HIV-1-infected patients in Liaoning Province. Methods Genomic DNA containing the proviral DNA of HIV 1 was extracted and amplified by nested PCR. The sequences were aligned, translated and analyzed. Results Infection with HIV-1 in Liaoning province belonged to the four subtypes A, B ’, C and G. The virus V3 ring in AIDS patients showed the amino acid mutation associated with the T-tropic SI phenotype (R, 13 (S, T, N), 19 (V), 20 (Y) and 25 (29) were higher than asymptomatic group (P <0.05). In addition, GQGR, APGQ , RPGA, GLGR, RPGA rare V3 ring top tetrapeptide composition and the fifth H, 34 S, F and other rare forms of mutation. Conclusions Amino acid mutations at the V3 loop of HIV-1 strains of A, B, C, and G subtypes in Liaoning Province among AIDS patients are generally similar to those of foreign subtype B strains. However, some rare and rare mutations were found V3 loop top tetrapeptide composition.
