目的:观察白藜芦醇对人白血病K562细胞的增殖与凋亡作用,及其调节K562细胞血管紧张素Ⅱ与血管内皮细胞生长因子的分泌,探讨其抗白血病机制。方法:人白血病K562细胞暴露于一定浓度梯度的白藜芦醇(12.5,25,50,100,200μmol·L-1),培养不同时间(12,24,36,48h)后,用细胞计数试剂盒-8法检测K562细胞增殖,流式细胞技术分析细胞细胞凋亡与细胞周期,采用放射免疫法检测细胞培养上清液中血管紧张素Ⅱ浓度,用酶联免疫吸附试剂检测细胞培养上清液中血管内皮生长因子的浓度。结果:白藜芦醇呈时间与浓度依赖性抑制K562细胞增殖并促进细胞凋亡;白藜芦醇处理K562细胞,培养细胞上清液中血管紧张素Ⅱ与血管内皮生长因子含量明显降低,与对照组相比,差异有统计学意义。结论:白藜芦醇通过下调血管紧张素Ⅱ与血管内皮生长因子的分泌而抑制K562白血病细胞增殖,诱导白血病细胞凋亡。
Objective: To observe the effect of resveratrol on the proliferation and apoptosis of human leukemia K562 cells, and to regulate the secretion of angiotensin II and vascular endothelial growth factor from K562 cells, and to explore its anti-leukemia mechanism. METHODS: Human leukemia K562 cells were exposed to a certain concentration gradient of resveratrol (12.5, 25, 50, 100, 200 μmol·L-1), cultured for different times (12, 24, 36, 48 h), and then used for cell counting kit-8 The proliferation of K562 cells was detected by flow cytometry, and cell apoptosis and cell cycle were analyzed by flow cytometry. The concentration of angiotensin II in cell culture supernatant was measured by radioimmunoassay, and the blood vessel in the supernatant of cell culture was detected by enzyme-linked immunosorbent assay. The concentration of endothelial growth factor. RESULTS: Resveratrol inhibited the proliferation of K562 cells and promoted cell apoptosis in a time and concentration-dependent manner. The resveratrol treatment of K562 cells significantly decreased the contents of angiotensin II and vascular endothelial growth factor in culture supernatants. Compared with the control group, the difference was statistically significant. CONCLUSION: Resveratrol can inhibit the proliferation of K562 leukemia cells and induce apoptosis of leukemia cells by down-regulating the secretion of angiotensin II and vascular endothelial growth factor.
