目的 探讨APP1 7肽是否能提高去卵巢大鼠学习、记忆功能及作用机制。方法 雌性大鼠分三组 :①正常组 ,②去卵巢组 ,③用APP1 7肽保护去卵巢组。通过水迷宫测试 ,观察行为学的改变。而后灌注固定 ,取大脑组织冰冻切片 ,做NT 3、NF免疫组化染色。结果 (1 )行为学检查 :给予APP1 7肽的卵巢去势大鼠游完全程的时间及错误反应次数均较未给予APP1 7肽的卵巢去势大鼠少 (P<0 0 5)。 (2 )用APP1 7肽保护的卵巢去势大鼠海马区神经元神经营养素 3(NT 3)、神经丝蛋白 (NF)的表达与正常大鼠相似 ,阳性细胞计数及平均灰度 (反映染色强度 )与卵巢去势大鼠比较差异显著 (P <0 0 5)。结论 卵巢去势大鼠存在学习、记忆障碍 ,APP1 7肽具有防治因雌激素缺乏而致神经元退变的功效
Objective To investigate whether APP1 7 peptide can improve the learning and memory function and the mechanism of action in ovariectomized rats. Methods Female rats were divided into three groups: ① normal group, ② ovariectomized group, ③ protected ovarian group with APP1 7 peptide. Pass the water maze test to observe behavioral changes. Then perfusion fixed, take frozen sections of brain tissue, so NT 3, NF immunohistochemical staining. Results (1) Behavioral examination: The ovary castration rats given APP1 7 peptide had less complete time and error reaction than those ovariectomized rats without APP1 7 peptide (P <0.05). (2) The expression of neurotrophin 3 (NT 3) and neurofilament (NF) in hippocampal neurons protected by APP1 7 peptide in ovariectomized rats were similar to those in normal rats. The positive cell counts and the average grayscale Intensity) compared with ovariectomized rats significant difference (P <0 05). Conclusion Ovariectomized rats have learning and memory impairment. APP1 7 peptide has the effect of preventing and treating neuronal degeneration caused by estrogen deficiency
