大黄提取物制备纳米银抑菌材料形态及抑菌性能研究

目的:探究大黄提取物制备纳米银抑菌材料的形态及其抑菌性能研究。方法:以大黄提取物为还原剂,使用生物还原法制备纳米银颗粒,分别通过紫外可见光光谱扫描、动态光散射及抑菌杀菌试验,观察不同制备方法下纳米银颗粒形态、稳定性及抑菌杀菌能力。结果:空白纳米银及供试品特征吸收峰为425 nm,在500~700nm波长范围内,无特征吸收峰出现,两种方法制备纳米银颗粒均未出现团聚体。大黄提取物制备的纳米银颗粒粒径为40.3 nm,较空白纳米银颗粒(52.1 nm)小。伴随加速实验时间延长,供试品及空白纳米银颗粒大小均呈现增大趋势。60℃加速35 d后空白纳米银颗粒粒径为392.7 nm;供试品粒径为68.1 nm,增幅小于空白对照组,表明大黄提取物制备纳米银颗粒有助于增加纳米银颗粒稳定性,避免纳米银颗粒过度聚集。大黄提取物能大大提高了纳米银的抗菌能力,大肠埃希菌最小抑菌浓度为50μg/m L,较空白纳米银颗粒抑菌活性增加了10倍;而金黄色葡萄球菌最小抑菌浓度只需50μg/m L,较空白纳米银颗粒抑菌活性增加了30倍。对上述两种细菌,最小杀菌浓度分别提高了4倍及20倍。结论:大黄提取物不仅能够提高纳米银颗粒的稳定性,还能增强纳米银颗粒抑菌活性。 Objective: To investigate the morphology and antibacterial activity of nano-silver antibacterial materials prepared from rhubarb extract. Methods: Rhodiola extract was used as reductant to prepare nano-silver particles by biological reduction method. The morphology, stability and antibacterial activity of nano-silver particles under different preparation methods were observed by UV-visible spectrum scanning, dynamic light scattering Bactericidal ability. Results: The characteristic absorption peak of the blank nano-silver and the test sample was 425 nm. There was no characteristic absorption peak in the wavelength range of 500-700 nm. No agglomeration occurred in the two methods. Rhubarb extract prepared nano-silver particle size of 40.3 nm, smaller than the blank nano-silver particles (52.1 nm). Accompanying the accelerated experimental time, the size of the sample and blank nano-silver particles showed an increasing trend. After accelerated at 60 ℃ for 35 d, the size of the blank silver nanoparticles was 392.7 nm. The diameter of the sample was 68.1 nm, which was smaller than that of the blank control group, indicating that the preparation of nano-silver particles by rhubarb extract can help to increase the stability of nano-silver particles and avoid Nano silver particles excessive accumulation. Rhubarb extract can greatly enhance the antibacterial capacity of nano-silver, Escherichia coli minimum inhibitory concentration of 50μg / m L, compared with blank nano-silver particles antibacterial activity increased 10-fold; and Staphylococcus aureus minimal inhibitory concentration only Need 50μg / m L, compared with blank nano-silver particles antibacterial activity increased by 30 times. For the two bacteria, the minimum bactericidal concentration increased by 4 times and 20 times. Conclusion: Rhubarb extract can not only improve the stability of nano silver particles, but also enhance the antibacterial activity of nano silver particles.
