目的探讨中药治疗前后结肠炎小鼠Foxp3基因和白介素-1β及诱导型一氧化氮的表达规律及与结肠炎发病的关系。方法建立二硝基氯苯+醋酸复合法诱发的小鼠溃疡性结肠炎模型,设立正常对照、模型对照、柳氮磺氨吡啶和中药灌肠1号治疗共4组,用RT-PCR方法检测小鼠结肠组织Foxp3的表达,免疫组化方法检测白介素-1β及诱导型一氧化氮的变化规律。结果建模后1~2周,与CD4+CD25+Tregs细胞亚群密切相关的Foxp3的表达显著低于对照组,而促炎因子白介素-1β及诱导型一氧化氮的含量明显强于对照组;而应用SASP和中药灌肠治疗后Foxp3表达显著高于模型组,两种促炎因子的含量明显降低。结论中药灌肠1号通过调控Foxp3表达进而增加Tregs细胞数量和抑制促炎因子表达,发挥对溃疡性结肠炎的治疗作用。
Objective To investigate the expression of Foxp3, interleukin-1β and inducible nitric oxide in colitis mouse before and after treatment with traditional Chinese medicine and its relationship with the pathogenesis of colitis. Methods The model of ulcerative colitis induced by dinitrochlorobenzene + acetic acid complex was established. Conclusion Enema 1 can regulate the expression of Foxp3, increase the number of Tregs cells and inhibit the expression of proinflammatory cytokines and play a therapeutic role in ulcerative colitis.
