目的通过检测Cyclin D1和k-Ras在蒙古族和汉族宫颈癌患者肿瘤组织中的表达水平,探讨其发病机制,为宫颈癌诊断提供理论依据。方法采用免疫组织化学法检测45例宫颈癌患者(蒙古族20例,汉族25例),25例蒙古族正常宫颈女性,10例蒙古族宫颈上皮内瘤变(CIN)Ⅰ级患者,9例CINⅡ级患者,8例CINⅢ级患者中的Cyclin D1和k-Ras表达情况,分析Cyclin D1和k-Ras与蒙古族和汉族女性宫颈疾病发生、发展的关系。结果汉族与蒙古族宫颈癌患者k-Ras和Cyclin D1蛋白阳性表达率比较差异无统计学意义(P>0.05)。与蒙古族正常宫颈女性比较,蒙古族宫颈癌患者k-Ras和Cyclin D1蛋白阳性表达率明显增高(P<0.05),且随着临床分期的增加,其阳性表达率也增加(P<0.05);不同年龄、民族、病理类型的宫颈疾病患者Cyclin D1和k-Ras蛋白阳性表达率比较差异无统计学意义(P>0.05);不同组织学分型、淋巴结转移与否患者Cyclin D1和k-Ras蛋白阳性表达率比较差异有统计学意义,且Cyclin D1和k-Ras阳性表达率呈正相关。结论蒙古族宫颈癌患者k-Ras和Cyclin D1蛋白的高表达与恶性程度及预后有关。蒙古族宫颈癌患者中Cyclin D1和k-Ras的表达率较高且有相关性。
Objective To detect the expression of Cyclin D1 and k-Ras in the tumor tissues of Mongolian and Han patients with cervical cancer and to explore the pathogenesis of this disease and provide a theoretical basis for the diagnosis of cervical cancer. Methods Immunohistochemistry was used to detect 45 cases of cervical cancer (20 cases of Mongolian and 25 cases of Han), 25 cases of normal female cervical Mongolian, 10 cases of Mongolian cervical intraepithelial neoplasia (CIN) Ⅰ level, 9 cases of CIN Ⅱ And 8 cases of CIN Ⅲ patients Cyclin D1 and k-Ras expression, analysis of Cyclin D1 and k-Ras and Mongolian and Han women cervical disease occurrence and development. Results The positive expression rate of k-Ras and Cyclin D1 in Han and Mongolian patients with cervical cancer showed no significant difference (P> 0.05). Compared with normal Mongolian women with cervical cancer, the positive rates of k-Ras and Cyclin D1 in Mongolian patients with cervical cancer were significantly higher (P <0.05), and the positive rates were also increased with the clinical stage (P <0.05) ; There was no significant difference in the expression rates of Cyclin D1 and k-Ras between cervical cancer patients of different ages, ethnic groups and pathological types (P> 0.05); Cyclin D1 and k-Ras in patients with different histological types and lymph node metastasis The positive rate of protein expression was significantly different, and the positive rate of Cyclin D1 and k-Ras was positively correlated. Conclusion The high expression of k-Ras and Cyclin D1 in Mongolian patients with cervical cancer is related to the degree of malignancy and prognosis. The expression of Cyclin D1 and k-Ras in Mongolian patients with cervical cancer was higher and correlated.
