目的评价131I-抗VEGF单克隆抗体(Sc-7269)瘤内注射放射免疫治疗荷人骨肉瘤裸鼠的疗效。方法24只荷人骨肉瘤裸鼠分别自瘤内注射(n=12)或静脉注射(n=12)131I-Sc-7269,进行生物分布研究。另20只分为4组,分别予瘤内注射等体积的生理盐水、11.1MBq的Na131I溶液、20g的Sc-269及11.1MBq的131I-Sc-7269。治疗后观察肿瘤体积2倍增时间及检测外周血白细胞计数。结果局部给药后肿瘤组织的放射性摄取在各个时间点明显高于系统给药,而其他组织的放射性摄取明显低于系统给药组。131I-Sc-7269治疗组裸鼠肿瘤体积2倍增时间明显延长(16.8±3.4/d)与其他各组相比有显著性差异(P<0.01),而该组治疗后外周血白细胞计数虽有下降(3520±248/mm3)但与对照组相比无统计学意义。结论131I-Sc-7269瘤内注射放射免疫治疗荷人骨肉瘤裸鼠是安全、有效的。
Objective To evaluate the efficacy of 131I-anti-VEGF monoclonal antibody (Sc-7269) intratumoral injection of radioimmunotherapy in human osteosarcoma bearing nude mice. Methods Twenty-four human osteosarcoma nude mice were biodistribution studies with intratumoral injection (n = 12) or intravenous (n = 12) 131I-Sc-7269, respectively. The other 20 were divided into 4 groups, respectively, to intratumoral injection of equal volume of saline, 11.1MBq of Na131I solution, 20g of Sc-269 and 11.1MBq of 131I-Sc-7269. After treatment, the tumor volume doubling time and the detection of peripheral blood leukocyte count. Results The radioactivity uptake of tumor tissue after topical administration was significantly higher than that of systemic administration at various time points, while the radioactivity uptake of other tissues was significantly lower than that of systemic administration group. The doubling time of doubling tumor volume in 131I-Sc-7269 treatment group was significantly longer (16.8 ± 3.4 / d) than that in other groups (P <0.01), while the peripheral blood leukocyte count Decreased (3520 ± 248 / mm3) but not statistically significant compared with the control group. Conclusion 131I-Sc-7269 intratumoral injection of radioimmunotherapy in human osteosarcoma nude mice is safe and effective.
