目的 探讨基质金属蛋白酶 - 2、9(MMP- 2、9)及影响二者作用的转化生长因子β1 (TGF-β1 )在声带息肉中的表达及意义 ,进一步探讨其发病机制。 方法 (1 )应用苏木精 -伊红染色及 Grocott- zones PISM复合法基底膜染色观察声带息肉基底膜的改变 ,并与声带小结及正常声带标本作比较。(2 )采用免疫组织化学方法检测 MMP- 2 /MMP- 9、TGF- β1在声带息肉、声带小结、正常声带粘膜中的表达及分布情况。 结果 (1 )苏木精 -伊红染色及 Grocott- zones PISM复合法基底膜染色检查 ,与正常声带相比 ,声带息肉粘膜上皮下基底膜正常或变薄 ,但血管内皮下基底膜比正常声带血管内皮下基底膜薄、稀疏 (P<0 .0 5 ) ,且粘膜固有层 (L P)间质水肿 ,变性 ,血管扩张 ,血栓形成等。与正常声带、声带息肉相比 ,声带小结粘膜上皮下基底膜增厚 (P <0 .0 5 )且不规则 ,但血管内皮下基底膜正常 ,粘膜固有层结构不改变或改变较轻。(2 )免疫组织化学染色 ,MMP- 2、MMP- 9、TGF- β1 在声带息肉的表达程度明显高于声带小结 (P <0 .0 1 )及正常声带粘膜 (P<0 .0 5 ) ,MMP- 2、MMP- 9、TGF-β1 在声带小结表达的阳性率比正常声带粘膜的表达的阳性率低 ,但 P>0 .0 5。声带息肉组织中 MMP- 2、MMP- 9与 TGF-β1 阳性表达之间存在正
Objective To investigate the expression and significance of matrix metalloproteinase - 2, 9 and transforming growth factor β1 (TGF-β1) in vocal cord polyp and to explore its pathogenesis. Methods (1) Hematoxylin-eosin staining and Grocott-zones PISM composite staining were used to observe the change of vocal cord polyp basement membrane, and compared with vocal nodules and normal vocal cords. (2) The expression and distribution of MMP-2 / MMP-9 and TGF-β1 in vocal cord polyp, vocal nodules and normal vocal cord mucosa were detected by immunohistochemistry. Results (1) Hematoxylin-eosin staining and Grocott-zones PISM composite staining of basilar membrane showed that the submucosal basement membrane of vocal cord polyp was normal or thinner than that of normal vocal cord, but the vascular basement membrane was lower than that of normal vocal cord The vascular basement membrane was thin and sparse (P <0.05), and interstitial edema, degeneration, vasodilation and thrombosis in the lamina propria of the mucosa. Compared with normal vocal cord and vocal cord polyp, the submucosal basement membrane thickens (P <0.05) and is irregular, but the vascular basement membrane is normal and the lamina propria of the mucosa does not change or changes slightly. (2) Immunohistochemical staining revealed that the expression of MMP-2, MMP-9 and TGF-β1 in vocal cord polyp was significantly higher than that in vocal nodules (P <0.01) and normal vocal cord mucosa (P <0.05) The positive rate of MMP-2, MMP-9 and TGF-β1 expression in vocal nodules was lower than that in normal vocal cord mucosa, but P> 0.05. There was a positive correlation between the expression of MMP-2, MMP-9 and TGF-β1 in vocal cord polyps
