目的:探讨山药对卵巢切除所致大鼠骨质疏松症的治疗作用,并通过观察其对OB和MSC OPG、RANKL蛋白及其mRNA表达的影响,探索其作用机理。方法:将60只雌性Wistar大鼠随机分为5组,即空白对照组、假手术组、模型组、阳性对照组和山药组,每组12只。摘除大鼠双侧卵巢1个月后,灌胃给予山药水煎剂,共给药3个月。采用骨组织形态计量学的方法对大鼠胫骨不脱钙骨切片进行形态计量;采用免疫组化和原位杂交的方法分别检测OB和MSC OPG、RANKL蛋白及其mRNA的表达。结果:与模型组比较,山药组大鼠胫骨TBV%显著增高,TRS%以及TFS%、MAR、OSW和mAR均明显降低;同时OB和MSC OPG蛋白和mRNA表达皆显著增高,而RANKL蛋白和mRNA表达皆显著降低。结论:山药对卵巢切除所致的骨质疏松症具有明显的治疗作用,能促进OB和MSC OPG的表达并抑制RANKL的表达,是其能够治疗骨质疏松症的机理之一。
OBJECTIVE: To investigate the therapeutic effect of yam on osteoporosis induced by ovariectomy in rats and its mechanism of action by observing the effects of OPG and RANKL on the expression of OPG and RANKL in OB and MSC. Methods: Sixty female Wistar rats were randomly divided into five groups: blank control group, sham operation group, model group, positive control group and yam group, 12 rats in each group. After removal of bilateral ovary in rats for 1 month, yam decoction was administered orally for a total of 3 months. Morphometry of non-decalcified tibial bone sections was performed by bone histomorphometry. The expressions of OPG and RANKL protein and mRNA were detected by immunohistochemistry and in situ hybridization. Results: Compared with the model group, the TBV% of the tibia and the percentage of TFS%, MAR, OSW and mAR in the yam group were significantly decreased, while the expression of OPG protein and mRNA in OB and MSC were significantly increased, while the RANKL protein and mRNA Expression were significantly reduced. Conclusion: Yam has a significant therapeutic effect on osteoporosis induced by ovariectomy. It can promote the expression of OB and MSC OPG and inhibit the expression of RANKL, which is one of the mechanisms that can treat osteoporosis.
