目的:建立测定丹参各营养器官中丹参酮IIA的高效液相色谱分析法,并用该方法测定丹参各营养器官中丹参酮IIA含量。方法:采用反相高效液相色谱法,以Zorbax SB-C18(150mm×4.6mm,5.0m)为色谱柱,柱温为30℃;甲醇:水=75:25为流动相;流速:1.0mL.min-1;检测波长:268nm。结果:丹参酮IIA浓度在4.4~70.4μg.mL-1(r=0.9996)范围内线性关系良好,平均加样回收率(n=6)为99.7%(R S D=2.1%)。结论:该法是一种快速、灵敏、准确的分析方法,可以为丹参的种植、采收、品质鉴定、扩展药用部位以及控制丹参药材质量提供理论依据。
Objective: To establish a HPLC method for the determination of tanshinone II A in vegetative organs of Salvia miltiorrhiza, and to determine the content of tanshinone II A in various vegetative organs of Salvia miltiorrhiza. Methods: The reversed-phase high performance liquid chromatography was used. The chromatographic column was Zorbax SB-C18 (150mm×4.6mm, 5.0m). The column temperature was 30°C; the mobile phase was methanol:water=75:25; the flow rate was 1.0mL. .min-1; detection wavelength: 268 nm. Results: The tanshinone IIA concentration was linear in the range of 4.4-70.4μg.mL-1 (r=0.9996). The average recovery (n=6) was 99.7% (R S D=2.1%). Conclusion: This method is a rapid, sensitive and accurate analysis method. It can provide theoretical basis for planting, harvesting, quality identification, expanding medicinal sites and controlling the quality of Salvia miltiorrhiza.
