本文以感病小麦幼苗为材料,首次对植物材料的微波辐射固定技术进行了研究。结果发现,感病叶片与固定液一起被微波辐射10S,再在固定液中放置10min,即可完成前固定过程。该方法处理的样品,其超微结构的保存效果与常规固定法(3h)相似,但对菌丝体表抗原位点的保存效果是常规法的2倍。
In this paper, the susceptible wheat seedlings as the material, for the first time on the microwave radiation fixation of plant materials were studied. The results showed that the infected leaves and the fixed solution were microwave radiation 10S, and then placed in a fixed solution for 10min, to complete the pre-fixation process. The sample treated by this method has the same effect of preserving the ultrastructure as conventional immobilization method (3h), but preserves the antigenicity of the mycelial surface at twice that of the conventional method.
