目的研究中国汉族人细胞色素P450 2J2(CYP2J2)基因启动子区CYP2J2*7(G-50T)多态性分布频率及其与高血压和脑卒中的相关性。方法采用聚合酶链反应-限制性内切酶片段长度多态(PCR-RFLP)方法,对260例脑卒中患者、364例原发性高血压(EH)患者及年龄、性别与之相近的243例健康对照者进行CYP2J2*7多态性分析。结果脑卒中组、EH组及正常对照组CYP2J2*1(野生型)等位基因频率分别为0.963,0.962和0.957;CYP2J2*7等位基因频率分别为0.037,0.038和0.043。CYP2J2*7等位基因频率与高血压和脑卒中无明显相关性(P>0.05)。三组之间3种等位基因型分布频率无明显差异(均P>0.05),其中CYP2J2*1三组分别为92.69%、92.86%和91.36%;CYP2J2*7/*1分别为6.92%、6.87%和8.64%;CYP2J2*7分别为0.38%、0.27%和0。结论CYP2J2*7等位基因频率与高血压和脑卒中无明显相关性。
Objective To investigate the distribution of CYP2J2 * 7 (G-50T) polymorphism in Chinese Han population and its relationship with hypertension and stroke. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used in 260 cases of stroke patients, 364 cases of essential hypertension (EH) and age and gender similar to 243 Cases of healthy controls CYP2J2 * 7 polymorphism. Results The allele frequencies of CYP2J2 * 1 (wild type) in stroke group, EH group and normal control group were 0.963, 0.962 and 0.957, respectively. The frequencies of CYP2J2 * 7 alleles were 0.037, 0.038 and 0.043, respectively. CYP2J2 * 7 allele frequency and no significant correlation between hypertension and stroke (P> 0.05). There was no significant difference in the frequencies of the three alleles between the three groups (all P> 0.05), among which CYP2J2 * 1 was 92.69%, 92.86% and 91.36% respectively; CYP2J2 * 7 / * 1 was 6.92% 6.87% and 8.64% respectively; CYP2J2 * 7 was 0.38%, 0.27% and 0 respectively. Conclusion There is no significant correlation between CYP2J2 * 7 allele frequency and hypertension and stroke.
