目的建立超高效液相色谱(UPLC)法同时测定豆芽中4种植物生长调节剂6-苄基腺嘌呤(6-BAP)、异戊烯基腺嘌呤(z-IP)、4-氟苯氧乙酸(4-FPA)、4-氯苯氧乙酸(4-CPA)的方法。方法样品采用乙腈提取,盐析离心,上清液过PXC/PWA固相萃取小柱净化,经C18色谱柱(100 mm×2.1 mm,1.8μm)分离,乙腈和磷酸二氢钠溶液梯度洗脱,二极管阵列检测器检测,外标法定量。结果 6-BAP和z-IP在0.25~25μg/mL范围内,4-FPA和4-CPA在0.50~50μg/mL范围内,线性关系良好,相关系数均>0.999;对4种植物生长调节剂进行低、中、高(6-BAP和z-IP:0.05、5.0和10.0 mg/kg;4-FPA:0.25、10.0和20.0 mg/kg;4-CPA:0.05、10.0和20.0 mg/kg)三浓度水平加标,回收率为70.0%~96.4%,相对标准偏差(RSD)为2.84%~12.10%(n=6)。检出限为0.0082~0.075 mg/kg,定量限为0.027~0.25 mg/kg。结论该法采用固相萃取进行样品前处理,简便、快捷,UPLC同时检测豆芽中6-BAP、z-IP、4-FPA和4-CPA,具有较好的准确度、灵敏度和精密度。
OBJECTIVE To establish a method for the simultaneous determination of 4 plant growth regulators, 6-benzyladenine (6-BAP), isopentenyladenine (z-IP), 4-fluorophenoxy Acetic acid (4-FPA), 4-chlorophenoxyacetic acid (4-CPA). Methods The samples were extracted with acetonitrile, salted out by centrifugation, the supernatant was purified by PXC / PWA solid phase extraction cartridges, separated on a C18 column (100 mm × 2.1 mm, 1.8 μm) and eluted with acetonitrile and sodium dihydrogen phosphate gradient , Diode array detector detection, external standard quantitative. Results The linear relationship between 6-BAP and z-IP in the range of 0.25-25μg / mL and 4-FPA and 4-CPA in the range of 0.50-50μg / mL were good with the correlation coefficients> 0.999. The effects of 4 plant growth regulators (6-BAP and z-IP: 0.05, 5.0 and 10.0 mg / kg; 4-FPA: 0.25, 10.0 and 20.0 mg / kg; 4-CPA: 0.05, 10.0 and 20.0 mg / kg) The results showed that the RSDs ranged from 2.84% to 12.10% (n = 6), with the recoveries of 70.0% ~ 96.4%. The detection limit was 0.0082 ~ 0.075 mg / kg, the limit of quantification was 0.027 ~ 0.25 mg / kg. Conclusions This method is simple, rapid and convenient. UPLC also has good accuracy, sensitivity and precision for simultaneous determination of 6-BAP, z-IP, 4-FPA and 4-CPA in bean sprouts.
