目的探讨子痫前期患者胎盘组织中线粒体解偶联蛋白2(UCP2)基因mRNA表达与胰岛素抵抗的关系。方法检测36例正常足月妊娠妇女(A组)和24例轻度子痫前期患者(B组)及17例重度子痫前期患者(C组)剖宫产前空腹血及新生儿脐血中血糖、胰岛素水平,采用RT-PCR法测定胎盘组织中UCP2 mRNA表达水平。结果C组新生儿脐血血糖和胰岛素显著低于A组(P<0.05);B、C组UCP2基因mRNA表达水平分别为(0.942±0.202)、(0.908±0.188),显著低于A组的(1.908±0.283)(P<0.05)。结论子痫前期患者胎盘组织中UCP2的低表达在子痫前期的发病中有重要作用,可能参与了胰岛素抵抗的病理生理过程。
Objective To investigate the relationship between mitochondrial uncoupling protein 2 (UCP2) gene mRNA expression and insulin resistance in placenta of preeclampsia. Methods 36 cases of normal term pregnant women (group A) and 24 cases of mild preeclampsia (group B) and 17 cases of severe preeclampsia (group C) cesarean section fasting blood and neonatal umbilical cord blood Blood glucose and insulin levels. UCP2 mRNA expression in placenta was determined by RT-PCR. Results The neonatal cord blood glucose and insulin in group C were significantly lower than those in group A (P <0.05). The mRNA expressions of UCP2 in group B and C were (0.942 ± 0.202) and (0.908 ± 0.188), respectively, which were significantly lower than those in group A (1.908 ± 0.283) (P <0.05). Conclusion The low expression of UCP2 in placental tissue of preeclampsia plays an important role in the pathogenesis of preeclampsia and may be involved in the pathophysiological process of insulin resistance.
