目的:探讨血浆microRNA(miRNA)在异基因造血干细胞移植(allo-HSCT)后发生急性移植物抗宿主病(aGVHD)与未发生a GVHD(non-a GVHD)患者的表达情况。方法:采用实时定量PCR(real-time PCR)的方法检测25例患者allo-HSCT移植前和移植后外周血血浆中miR-423,miR199a-3p,miR-93*和miR-377的表达水平。结果:miR-423,miR199a-3p,miR-93*在a GVHD患者中的表达量较未发生a GVHD的患者高(P<0.05),而miR-377在两组之间表达无统计学差异(P=0.0818)。结论:异基因造血干细胞移植后发生a GVHD患者的血浆中miR-423,miR199a-3p,miR-93*表达增高,这可作为为监测和诊断a GVHD的生物标志物。
Objective: To investigate the expression of plasma microRNA (miRNA) in patients with acute graft-versus-host disease (aGVHD) and non-aGVHD (all-HSCT) after allogeneic hematopoietic stem cell transplantation (allo-HSCT). Methods: The expression of miR-423, miR199a-3p, miR-93 * and miR-377 in peripheral blood of 25 patients with allo-HSCT before and after transplantation were detected by real-time PCR. Results: The expression levels of miR-423, miR199a-3p and miR-93 * in patients with a GVHD were significantly higher than those without a GVHD (P <0.05), while the expression of miR-377 was not significantly different between the two groups (P = 0.0818). Conclusion: The expression of miR-423, miR199a-3p and miR-93 * in the plasma of patients with a GVHD after allogeneic hematopoietic stem cell transplantation is increased, which may serve as a biomarker for monitoring and diagnosing a GVHD.
