目的:通过高脂诱导营养性肥胖大鼠模型,观察调脂积冲剂对肥胖大鼠体重、Lee’s指数、血清胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、血糖(FBG)、真胰岛素(FTI)、胰岛素抵抗指数(HOMA-IR)及游离脂肪酸(FFA)、脂联素、抵抗素的影响。方法:将SD雄性大鼠65只,分为正常组10只,其余55只采用高脂饮食建立肥胖模型。7周末时测体重,大鼠体重大于正常组体重20%为造模成功。将造模成功的30只大鼠随机分为模型组、调脂积组、荷丹片组。灌胃给药5周后,测量各组大鼠体质量、身长,计算Lee’s指数。检测各组大鼠血清TC、TG、HDL、LDL、FBG、FFA。采用ELISA法测定血清的真胰岛素、脂联素、抵抗素含量。采用稳态模式评估法(HOMA)来评价胰岛B细胞功能,即胰岛素抵抗指数(HOMA-IR)=(FBG)*FTI/22.5。结果:和正常组比较,模型组大鼠体重、Lee’s指数、TC、TG、LDL、FTI、HOMA-IR、FFA、抵抗素明显升高(P<0.01);FBG有差异性升高(P<0.05);HDL有差异性降低(P<0.05);脂联素明显降低(P<0.01)。和模型组比较,调脂积组大鼠体重明显下降(P<0.01);Lee’s指数、TG有差异性下降(P<0.05);TC、LDL明显下降(P<0.01);FBG略有下降、HDL略有升高,但无统计学意义(P>0.05);HOMA-IR、FTI、FFA明显下降(P<0.01);血清脂联素有差异性升高(P<0.05);血清抵抗素有差异性下降(P<0.05)。调脂积组和荷丹片组比较,在上述指标方面差异无统计学意义(P>0.05)。结论:高脂饲料诱导的肥胖大鼠体内存在胰岛素抵抗,血清FFA、抵抗素水平升高,脂联素水平降低;调脂积冲剂可以减轻肥胖大鼠的体重,调节血脂紊乱,改善其胰岛素敏感性,这种作用可能和降低血清FFA、抵抗素水平及升高脂联素水平有关。
OBJECTIVE: To observe the effects of Tiaozhi JI on body weight, Lee’s index, serum cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) LDL, FBG, FTI, HOMA-IR, free fatty acid (FFA), adiponectin and resistin. Methods: Sixty-five male Sprague-Dawley rats were divided into normal group (n = 10) and the remaining 55 were given a high-fat diet to establish an obesity model. 7 weighed at the end of the test weight, body weight greater than the normal 20% of the body weight for the success of modeling. Thirty rats with successful model were randomly divided into model group, lipid regulating group and holin tablets group. After gavage for 5 weeks, the body weight and length of the rats in each group were measured, and Lee’s index was calculated. Serum TC, TG, HDL, LDL, FBG and FFA were detected in each group. Serum real insulin, adiponectin and resistin were measured by ELISA. Steady-state pattern assessment (HOMA) was used to assess islet B-cell function, ie, insulin resistance index (HOMA-IR) = (FBG) * FTI / 22.5. Results: The body weight, Lee’s index, TC, TG, LDL, FTI, HOMA-IR, FFA and resistin in the model group were significantly higher than those in the normal group (P < 0.05), HDL decreased significantly (P <0.05), adiponectin decreased significantly (P <0.01). Compared with the model group, the body weight of the rats in the lipid regulating group decreased significantly (P <0.01); the Lee’s index and TG decreased significantly (P <0.05); TC and LDL decreased significantly (P <0.01) HDL increased slightly, but no statistical significance (P> 0.05); HOMA-IR, FTI, FFA decreased significantly (P <0.01); serum adiponectin increased significantly (P <0.05); serum resistin There are differences (P <0.05). There was no significant difference in the above indexes between the lipid regulating group and Halodan tablet group (P> 0.05). Conclusion: The obesity rats induced by high-fat diet have insulin resistance, the serum FFA and resistin levels are increased, and the adiponectin level is decreased. Tiaozhi Jie granules can reduce the weight of obese rats, regulate the dyslipidemia and improve their insulin sensitivity Sex, this effect may be related to lower serum FFA, resistin levels and increased adiponectin levels.
