应用阳性选择策略,以新型IsolexTM50系统建立了富集CD34+造血干/祖细胞(HSC/HPC)的免疫磁珠无菌分选术,对正常骨髓单个核细胞中的CD34+HSC/HPC进行富集与鉴定。结果表明,富集前CD34+HSC/HPC为1.4%~3.4%,富集后纯度高达87.5%~95.0%,回收率为49%~80%,仍保持细胞活力。该系统具有高效、快速和可复性好的特点,每次能分选2×109单个核细胞,可获取较多的CD34+HSC/HPC,整个分离过程≤3.5小时,为深入研究人类HSC/HPC的生物学特征与功能奠定了基础
The positive selection strategy was used to establish the immunomagnetic separation of CD34 + hematopoietic stem / progenitor cells (HSC / HPC) by the new IsolexTM50 system, and the enrichment and identification of CD34 + HSC / HPC in normal bone marrow mononuclear cells . The results showed that pre-enrichment CD34 + HSC / HPC was 1.4% ~ 3.4%, the enrichment purity was as high as 87.5% ~ 95.0%, the recovery was 49% ~ 80%, still maintain cell viability.
