吸附和扩散特性是进行色谱过程的基础,本文测定了紫杉醇及其类似物三尖杉磷碱在多孔硅胶上的吸附及扩散特性,结果表明:紫杉醇和三尖杉磷碱在硅胶上的吸附不符合Langmuir单分子层吸附理论,但在低浓度下可用溶剂调整型Langmuir模型描述。扩散研究表明;紫杉醇和三尖杉磷碱在硅胶上吸附较弱,表面扩散可以忽略。相同条件下,紫杉醇的吸附量受强溶剂影响较大,强溶剂组成的增加会造成紫杉醇吸附量很快下降.这和在正相色谱中紫杉醇先于三尖杉磷碱流出色谱柱的事实是相符的。
Adsorption and diffusion characteristics are the basis of the chromatographic process. The adsorption and diffusion characteristics of paclitaxel and its analogues, harringtonine, on porous silica gel were determined. The results showed that the adsorption of paclitaxel and cephalosporin on silica gel was not. In accordance with the Langmuir monolayer adsorption theory, it can be described by a Langmuir model with solvent adjustment at low concentrations. Diffusion studies have shown that the adsorption of paclitaxel and harringtonine on silica gel is weak and the surface diffusion is negligible. Under the same conditions, the adsorption amount of paclitaxel is greatly affected by the strong solvent, and the increase of the strong solvent composition will cause the decrease of paclitaxel adsorption. This is consistent with the fact that paclitaxel is eluted from the chromatographic column prior to harringtonine in normal phase chromatography.
