目的:从血液流变学和红细胞膜胆固醇的变化探讨丹参注射液改善老龄大鼠红细胞变形性的作用机制。方法:70~72周龄Wistar大鼠30只,雌雄各半,随机分为丹参注射液高剂量组(4.4 g生药/kg)、低剂量组(1.1 g生药/kg)和模型组。另外10只轻龄大鼠作为轻龄对照组,ip给药14 d后,取血测定全血黏度、血浆黏度和红细胞变性指数等指标,并用高效液相色谱法测定红细胞膜胆固醇含量。结果:与模型组相比,丹参注射液组能够显著降低老龄大鼠的全血黏度(P<0.01)、血浆黏度(P<0.05)以及红细胞膜上胆固醇含量(P<0.01);并能明显提高红细胞变形能力(P<0.01)。结论:丹参注射液可能通过减少红细胞膜上的胆固醇含量,影响红细胞膜上脂质代谢,是提高红细胞膜变形性,改善血液黏度的机制之一。
OBJECTIVE: To explore the mechanism of Salvia miltiorrhiza injection in improving erythrocyte deformability in aged rats from the changes of hemorheology and erythrocyte membrane cholesterol. Methods: Thirty Wistar rats, aged 70-72 weeks old, were randomly divided into three groups: high dose Salvia miltiorrhiza injection group (4.4 g crude drug / kg), low dose group (1.1 g crude drug / kg) and model group. Another 10 light-aged rats as a light-aged control group, 14 d after ip administration, blood samples were collected for determination of whole blood viscosity, plasma viscosity and erythrocyte degeneration index and other parameters. The contents of erythrocyte membrane cholesterol were determined by HPLC. Results: Compared with the model group, Salvia miltiorrhiza injection group could significantly reduce the whole blood viscosity (P <0.01), plasma viscosity (P <0.05) and the cholesterol content of erythrocyte membrane in aged rats (P <0.01) Improve erythrocyte deformability (P <0.01). Conclusion: Salvia miltiorrhiza injection may be one of the mechanisms of improving erythrocyte membrane deformability and improving blood viscosity by reducing cholesterol content in erythrocyte membrane and affecting lipid metabolism on erythrocyte membrane.
