目的探讨人重组粒细胞集落刺激因子(rhG-CSF)对大鼠缺血性脑梗死的治疗作用及其应用的时间窗。方法采用线栓法制作大鼠缺血性脑梗死模型,分别于梗死后1h、6h给予皮下注射rhG-CSF(50μg/kg);对照组给予同等剂量的生理盐水皮下注射。TTC染色检测梗死体积。结果术后48h处死大鼠,梗死后1h给药,治疗组与对照组梗死体积分别为(135.0±13.8)mm3、(230.0±57.8)mm3,两者比较P<0.001;梗死后6h给药,梗死体积分别为(140.0±25.3)mm3、(240.0±30.6)mm3,两者比较P<0.001。结论rhG-CSF可减轻大鼠缺血性脑梗死程度,减小梗死体积。术后6h给药与术后1h给药具有相当的疗效。
Objective To investigate the therapeutic effect of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on ischemic cerebral infarction in rats and the time window of its application. Methods The rat model of ischemic cerebral infarction was established by the method of thread plug. The rhG-CSF (50μg / kg) was given subcutaneously at 1 hour and 6 hours after infarction. The control group was injected subcutaneously with the same dose of saline. TTC staining was used to measure infarct volume. Results The rats were sacrificed 48h after operation and 1h after infarction. The volumes of infarction in the treatment group and control group were (135.0 ± 13.8) mm3 and (230.0 ± 57.8) mm3, respectively, P <0.001; The infarct volumes were (140.0 ± 25.3) mm3 and (240.0 ± 30.6) mm3, respectively, both P <0.001. Conclusion rhG-CSF can reduce the degree of ischemic cerebral infarction and decrease the infarct volume in rats. Postoperative 6h administration and postoperative 1h administration has a considerable effect.
